XPC Antibody (D-10) is a mouse monoclonal IgG2a kappa light chain antibody that detects XPC protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-XPC antibody (D-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. XPC protein plays a crucial role in the nucleotide excision repair (NER) pathway, which maintains genomic stability by recognizing and repairing bulky DNA lesions, such as those caused by ultraviolet light. Deficiencies in this repair mechanism can lead to severe consequences, including an increased risk of skin cancers, particularly in individuals with Xeroderma Pigmentosum (XP), a genetic disorder characterized by extreme sensitivity to sunlight. XPC gene, located on chromosome 3p25, encodes a protein that forms a complex with Cen2 and HR23B, which are vital for stabilizing XPC and facilitating DNA damage recognition. The carboxy-terminus of XPC is critical for binding to HR23B and damaged DNA; mutations that truncate this region can result in nonfunctional XPC proteins, further underscoring XPC′s importance in the DNA repair process.

Has this antibody been tested in mouse cells for WB? It is a mouse monoclonal antibody, wouldn't it give a huge background if used for detecting mouse XPC?

Asked by: TKOK
Thank you for your question. This antibody is recommended and warrantied for use on mouse samples. Background issues are generally not an issue with proper conditions. You can find our recommended protocols here: https://www.scbt.com/scbt/resources/protocols

Does this antibody recognise ubiquitylated XPC?

Asked by: XPC_Query
Thank you for your question. This antibody will detect ubiquitylated XC as long as the ubiquitination site isn't where the epitope is. It is not specifically designed to detect the ubiquitylated form, so this is not a guarantee that it will pick it up. Please contact our Technical Service Department by phone, e-mail or live chat if you have any further questions.
