Wnt11 encodes the secreted ligand Wnt-11, a key modulator of non-canonical Wnt signaling that shapes cell polarity, directional migration, and tissue morphogenesis during development. Wnt-11 commonly signals through planar cell polarity and Wnt/Ca²⁺ pathways, engaging receptors such as Frizzled and ROR to influence cytoskeletal dynamics and calcium-dependent transcriptional programs. In mouse systems, Wnt-11 activity is linked to organogenesis processes including cardiac and craniofacial patterning, and it can intersect with β-catenin–dependent signaling in a context-specific manner. Dysregulated Wnt11 signaling has been associated with abnormal morphogenesis, altered cell motility programs, and phenotypes relevant to fibrosis and cancer biology models.
Wnt-11 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Wnt11 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Wnt11 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Wnt11 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Wnt-11 protein expression.
This CRISPR knockout system enables efficient generation of Wnt11-deficient cell models for investigation of Wnt-11 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
