WIP Antibody (C-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects WIP protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-WIP antibody (C-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The Wiskott-Aldrich syndrome protein (WASP) is crucial for immune function, and mutations in WASP can lead to immunodeficiency due to improper T cell and B cell activation. The 503 amino acid WAS-interacting protein (WIP) plays a significant role in this process, containing several domains that facilitate actin-binding and src homology interactions. Notably, the first 100 amino acids of WASP interact with amino acids 377-503 of WIP, and most pathogenic mutations associated with Wiskott-Aldrich syndrome occur within this region. The gene encoding human WIP is located on chromosome 2q31.1, and studies have shown that WIP overexpression in the human B cell line BJAB enhances F-actin content and promotes cerebriform projection formation. Additionally, WIP and Vav work together to regulate NF-AT/AP-1 gene transcription, with the WIP-WASP complex being essential for NF-AT/AP-1 activation, which is vital for proper T cell function. Disruption of the WIP-WASP complex may contribute to the immunodeficient phenotype observed in Wiskott-Aldrich syndrome, highlighting WIP′s importance in maintaining immune system integrity.
