WIF-1 Antibody (B-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects WIF-1 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). WIF-1 (B-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. WIF-1 plays a crucial role as a Wnt antagonist, which is essential for regulating the Wnt signaling pathway that governs numerous developmental processes such as embryogenesis and central nervous system patterning. By inhibiting Wnt signaling, WIF-1 ensures that the spatial and temporal expression of Wnt target genes is properly controlled, preventing aberrant cell proliferation and differentiation that could lead to developmental disorders or cancer. Structurally, WIF-1 is a 379-amino acid secreted protein characterized by an N-terminal signal sequence, a 150-amino acid WIF domain, five epidermal growth factor-like repeats, and a 45-amino acid hydrophilic C-terminal domain, which collectively contribute to binding Wnt proteins and modulating their activity. Anti-WIF-1 antibody (B-10) versatility in various applications makes this product an invaluable tool for researchers studying the intricate mechanisms of Wnt signaling and its implications in developmental biology and disease.

Hi, your representative western blot images show different molecular weight of WIF1 here. First one shows WIF1 band between 90k and 55k, and second one between 43k and 34k. It is almost 20k differences of band location between two blots. Why is it?

Asked by: BoomBoomPow
WIF-1 has a predicted molecular weight of 42 kDa. However, the observed endogenous molecular weight of WIF-1 in the cell lines tested is higher than the predicted molecular weight (~55-63 kDa) and is likely due to post-translational modifications such as glycosylation. The second Western blot image shown on our website is using a transfected lysate and is not necessarily representative of the endogenous molecular weight for WIF-1.

What application is the blocking peptide sc-373780 P appropriate for?

Asked by: cjMara
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization
