Mouse Hdlbp encodes vigilin, a large RNA-binding protein with multiple KH domains that associates with mRNA and ribonucleoprotein complexes to influence transcript stability, localization, and translation. Vigilin has been linked to regulation of lipid and sterol metabolic gene expression through interactions with specific mRNAs, connecting it to broader pathways controlling cellular energy balance and membrane homeostasis. It also participates in post-transcriptional gene control during proliferation and stress responses, consistent with roles in RNA processing and translational regulation. Dysregulation of vigilin-associated RNA networks has been investigated in contexts involving metabolic phenotypes and altered gene-expression programs relevant to disease mechanisms.
Vigilin CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Hdlbp gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Hdlbp together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Hdlbp open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Vigilin protein expression.
This CRISPR knockout system enables efficient generation of Hdlbp-deficient cell models for investigation of Vigilin signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
