VCAM-1 Antibody (E-10) is a mouse monoclonal IgG1 kappa light chain antibody raised against amino acids 25-300 of human Vascular Cell Adhesion Molecule 1 (VCAM-1). VCAM-1, also known as CD106, is a transmembrane glycoprotein belonging to the immunoglobulin superfamily and is primarily expressed on activated endothelial cells. Structurally, VCAM-1 comprises seven immunoglobulin-like domains essential for binding to integrins such as VLA-4 (α4β1) on leukocytes, facilitating their adhesion and transmigration across the endothelium during immune responses. Post-translational modifications like N-linked glycosylation are crucial for VCAM-1′s proper folding, stability, and function, influencing interaction with integrins and overall cell adhesion processes. VCAM-1 expression is upregulated by inflammatory cytokines such as interleukin-1 (IL-1), tumor necrosis factor-alpha (TNF-α), and lipopolysaccharide (LPS), linking VCAM-1 to various inflammatory conditions like atherosclerosis and rheumatoid arthritis.
