utrophin Antibody (8A4) is a mouse monoclonal IgG1 kappa light chain antibody that detects utrophin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. anti-utrophin antibody (8A4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Utrophin plays a crucial role in muscle function and cellular integrity, as utrophin is a structural, actin-binding protein that helps anchor the cytoskeleton to the plasma membrane, similar to dystrophin, which is associated with Duchenne and Becker muscular dystrophies. Utrophin′s importance lies in its ability to compensate for the loss of dystrophin in muscle tissues, particularly in muscular dystrophies. Research indicates that upregulating utrophin can mitigate the pathological effects of dystrophin deficiency, providing a potential therapeutic avenue for treating these debilitating conditions. Additionally, utrophin′s interaction with various proteins, including dystrophin-associated glycoproteins, highlights utrophin′s significance in maintaining muscle fiber structural integrity and facilitating communication between the cytoskeleton and extracellular matrix.

Thank you for your question. We do not have any recommended negative control cell lysates available for this antibody. Positive controls are C2C12 whole cell lysate: sc-364188, SJRH30 cell lysate: sc-2287 or Caco-2 cell lysate: sc-2262. Alternatively, negative controls can be achieved with our CRISPR- or si-/shRNA-based knockout/knockdown products. Have you ever used this antibody sc-33700 for mouse muscle utrophin WB, is there some unspecific bands?

Can you tell me the condition for Western blotting(or transfer condition) of UTRN? I tried to do western blotting of Utrophin, but I couldn't detect the specific band for utrophin. The reason is that utrophin is high molecular weight protein, I guess.

Hi, I was wondering whether the isotype is really IgG1. In the DSHB the same clone (Mancho3; 8A4) is described as IgG2a. I need to know whether I can establish a co-staining using isotype specific secondary abs. Can you confirm the isotype? Many thanks.

Thank you for your question. We have confirmed with our lab and according to internal isotype testing this antibody, utrophin (8A4): sc-33700 is a mouse monoclonal IgG1 (kappa light chain). This antibody is also available directly conjugated to HRP (sc-33700 HRP), phycoerythrin (sc-33700 PE), fluoroscein (sc-33700 FITC), Alexa Fluor® 488 (sc-33700 AF488) or Alexa Fluor® 647 (sc-33700 AF647), if you want to use the antibody in a multi-antibody staining application, and avoid any issues related to using a secondary antibody for detection.

could you tell me which amino acides Utrophin (8A4) against for, or which part of utrophin for example, C-terminal, N-terminal, or others of utrophin? could you write similar information like utrophin Sc-81557, see  data-sheet. utrophin (MANCHO7)

Thank you for your question. Upon further review in the literature, it does appear that this monoclonal antibody has been epitope mapped. This mouse monoclonal, utrophin Antibody (8A4): sc-33700, has an epitope mapping between amino acids 3232-3273 of human origin.

Can this antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Rated 5 out of 5 by from works fine in WB and IPwe were given a sample to try it out. It works fine in WB (final dilution 1:2000 to get 1ug/ml), for blocking I used 5% milk in TBST. It works fine in IP too, I used 1ug of AB and 2mg of lysate per IP. Since utrophin is a very high molecular weight protein, I used 6% SDS-PAGE gel to resolve it.
