SEE ALSO...

USP42 Antibody (D-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects USP42 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). USP42 (D-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. USP42 protein, which is 1,325 amino acids long and belongs to the peptidase C19 family, plays a crucial role in the ubiquitin-proteasome pathway, a vital cellular mechanism that regulates protein degradation and turnover. This pathway is essential for maintaining cellular homeostasis, as USP42 controls various intracellular processes such as signal transduction, transcriptional activation, and cell cycle progression. By catalyzing the conversion of a ubiquitin C-terminal thioester to free ubiquitin and a thiol, USP42 helps modulate ubiquitin levels available for tagging proteins for degradation, thereby influencing numerous cellular functions. USP42 gene is located on human chromosome 7, a region associated with several genetic disorders, including Osteogenesis imperfecta and Williams-Beuren syndrome, highlighting USP42′s importance in both normal physiology and disease states.

I am using this USP42 antibody. I loaded 20ug cell lysates to gels, transferred into PVDF membrane, blocking, incubated with anti-USP42 antibody (1:300 in 5% milk), 2nd Ab (1:5000). Detection time was 20min. How I can reduce exposure time to get signal?

Can USP42 (D-4): sc-390604 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: jenniferc
Thank you for your question. Yes, USP42 (D-4): sc-390604 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
