SEE ALSO...

USP34 Antibody (3H9) is a mouse monoclonal IgG2a kappa light chain antibody that detects USP34 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-USP34 antibody (3H9) is available as a non-conjugated format. USP34 protein consists of 3,395 amino acids and plays a crucial role in the ubiquitin-proteasome system, functioning as a deubiquitinating enzyme that removes ubiquitin moieties from target proteins. This activity is vital for regulating protein stability and function, particularly in response to cellular stressors such as DNA damage, where USP34 is phosphorylated by ATM or ATR kinases. By cleaving ubiquitin from both ubiquitinylated proteins and ubiquitin-fused precursors, USP34 prevents degradation by the proteasome, thereby maintaining essential cellular functions. USP34 is expressed at low levels in the brain and has two isoforms resulting from alternative splicing, which may contribute to diverse regulatory roles in various cellular processes. USP34′s ability to modulate the ubiquitin pathway underscores its importance in signal transduction, transcriptional regulation, and cell cycle control, making USP34 a significant target for research in cancer and other diseases where these processes are disrupted.

What percentage gel/ recommended transfer conditions do you suggest for a good band for USP34? I have used recommended dilution of 1:200 and see multiple bands (of similar intensity) above 250kd MW marker band.

Asked by: SMahajan
Thank you for your question. This is a very large protein, so we recommend a 8% gel for this protein. Please contact our Technical Service Department if you need further assistance with this antibody.

Can sc-100631: USP34 (3H9) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Trav11
Thank you for your question. Yes, sc-100631: USP34 (3H9) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
