uPAR Antibody (E-3) is a mouse monoclonal IgG2a kappa light chain antibody that detects uPAR protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). uPAR (E-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The urokinase plasminogen activator receptor (uPAR), also known as CD87, is a glycoprotein anchored to the cell surface and plays a critical role in regulating pericellular proteolysis. uPAR specifically binds to urokinase plasminogen activator (uPA), which catalyzes plasminogen conversion to plasmin, a serine protease essential for fibrin degradation in the extracellular matrix. This process is vital for various physiological functions, including tissue remodeling, wound healing, and inflammatory response. The interaction between uPA and uPAR is implicated in tumor biology, as both proteins are often overexpressed in cancer cells, facilitating tumor invasion and metastasis. The signaling cascade initiated by uPA binding to uPAR involves key downstream effectors such as Ras, MEK, ERK, and MLCK, which collectively promote cellular migration in a manner selective for integrins. This makes uPAR a significant target for therapeutic interventions aimed at inhibiting cancer progression and metastasis.

What is the recommended starting dilution for Western blot for uPAR (E-3): sc-376494?

Rated 1 out of 5 by from Very weak band in WBI use 293,HC67, Hela, SW1990,BXPC3,PANC1 cell lysis, delution 1:300.very weak band with a long exposure time. Besides, these bands almost similar.But many ref reported that 293 and HC67 do not express uPAR. Therefore,I think these weak bands are not uPAR. I wonder how these people commenting here get their results using this garbage antibody.

Rated 1 out of 5 by from No bands seenWestern blotting was performed using twelve different mouse osteosarcoma cell lines which were previously confirmed to express uPAR gene based on RT-PCR. Conditions: 12% gel electrophoresis, wet transfer over 2 hours, primary antibodies at 1:200 dilution in 5% mild blocking solution (4C overnight). There was no band visible following the protocol, although positive control (b-actin) was seen.
