UBE1 Antibody (D-8) is a mouse monoclonal IgG2a antibody that detects UBE1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). UBE1, or ubiquitin activating enzyme E1, plays a crucial role in the ubiquitin-proteasome pathway, which is essential for regulating protein degradation and maintaining cellular homeostasis. By catalyzing the first step in ubiquitin conjugation, UBE1 activates ubiquitin through an ATP-dependent process, forming a thiolester bond with a cysteine residue in UBE1 itself. This activated ubiquitin is then transferred to ubiquitin-conjugating enzymes, which subsequently attach ubiquitin to target substrates, marking them for degradation. The proper functioning of UBE1 is vital, as dysregulation of this pathway can lead to various diseases, including cancer and neurodegenerative disorders. Notably, the UBE1 gene is an example of an X-Y homologous gene, being X-linked with a distinct Y-linked counterpart in many mammals, although no homolog is found on the human Y chromosome. Interestingly, UBE1 is believed to escape X inactivation in humans, which may have implications for gene dosage and expression in females. Anti-UBE1 antibody (D-8) is an invaluable tool for researchers studying the dynamics of protein degradation and the intricate mechanisms of cellular regulation.
