UBE1 Antibody (2G2) is a mouse monoclonal IgG1 kappa light chain antibody that detects the UBE1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-UBE1 antibody (2G2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The ubiquitin activating enzyme E1 (UBE1) plays a crucial role in the ubiquitin-proteasome pathway, which is essential for regulating protein degradation and maintaining cellular homeostasis. UBE1 catalyzes the initial step of ubiquitin conjugation by adenylating the C-terminal glycine residue of ubiquitin in an ATP-dependent manner, forming a thiolester bond with a cysteine residue on UBE1. This activated ubiquitin is subsequently transferred to a ubiquitin-conjugating enzyme, which then donates the ubiquitin moiety to target substrates, marking them for degradation. Proper functioning of UBE1 is vital, as dysregulation of this pathway can lead to various diseases, including cancer and neurodegenerative disorders. Notably, the UBE1 gene is an example of an X-Y homologous gene, being X-linked with a distinct Y-linked counterpart in many mammals, although no UBE1 homolog is found on the human Y chromosome. UBE1 is believed to escape X inactivation in humans, which may have implications for gene dosage and expression in females.

Can UBE1 (2G2): sc-53555 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Asked by: jenniferc
Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, UBE1 (2G2): sc-53555 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
