Tnnt3 encodes fast skeletal muscle troponin T (Troponin T-FS), a core component of the thin filament troponin complex that couples cytosolic Ca²⁺ binding to tropomyosin movement and actin–myosin cross-bridge cycling. By anchoring the troponin complex to tropomyosin, Troponin T-FS helps set contraction kinetics and fiber-type–specific contractile performance during excitation–contraction coupling. Tnnt3 regulation intersects with myofibrillogenesis, sarcomere maintenance, and calcium-dependent signaling that tunes muscle adaptation to activity and load. Dysregulation or altered isoform usage of fast troponin T is relevant to studies of skeletal muscle weakness, myofibrillar disorganization, and related neuromuscular disease mechanisms.
Troponin T-FS CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Tnnt3 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Tnnt3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Tnnt3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Troponin T-FS protein expression.
This CRISPR knockout system enables efficient generation of Tnnt3-deficient cell models for investigation of Troponin T-FS signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
