TRK-T3 Antibody (H-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects TRK-T3 protein of human origin, TFG protein of mouse origin, and the corresponding rat homolog by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-TRK-T3 antibody (H-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TRK-T3 protein is a product of oncogenic rearrangements of the NTRK1 gene, which encodes the Trk A protein, and these rearrangements are frequently observed in thyroid carcinomas. The fusion of the NTRK1 tyrosine kinase domain with 5′-end sequences of different genes results in TRK-T3 oncoprotein, which consists of 1,412 nucleotides of NTRK1 preceded by 598 nucleotides from TFG (TRK-fused gene), a gene that is ubiquitously expressed and located on chromosome 3. TRK-T3 protein includes a coiled-coil motif within the TFG region, which is crucial for forming protein complexes. This structural feature enhances interactions with other signaling molecules, allowing cytoplasmic TRK-T3 protein to bind to and phosphorylate adaptor proteins such as Shc and SNT1/FRS2. These interactions are vital for linking receptor tyrosine kinases to the mitogen-activated protein kinase pathway, thereby facilitating cellular signaling processes that can lead to tumorigenesis. SHP-1 interacts with and down-regulates TRK-T3, highlighting the importance of post-translational regulation in maintaining cellular homeostasis.
