TRIM5 Antibody (D-6) is a mouse monoclonal IgG1 kappa light chain antibody that detects TRIM5 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TRIM5 (D-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TRIM5 protein, consisting of 493 amino acids, is a crucial member of the tripartite motif (TRIM) protein family, which is characterized by three zinc-binding domains: a RING domain, a B-box 2, and a coiled-coil domain. These structural features enable TRIM proteins to engage in homomultimerization, allowing them to localize to specific cellular compartments. Notably, TRIM5 also contains a carboxy-terminal B30.2 (SPRY) domain that is essential for mediating innate intracellular retroviral resistance. This domain′s three variable regions form loops that may serve as a binding surface for retroviruses, highlighting TRIM5′s importance in antiviral defense. The trimerization of isoform alpha of TRIM5 significantly enhances affinity for the retroviral capsid, thereby inhibiting viral infection after virus entry into the host cell. The linker region connecting the coiled-coil and B30.2 domains is critical for this trimerization process, underscoring the intricate relationship between TRIM5′s structure and functional capabilities in combating retroviral threats.
