TRIM32 Antibody (8H8) is a mouse monoclonal IgG2a kappa light chain antibody that detects TRIM32 protein of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). anti-TRIM32 antibody (8H8) is available as the non-conjugated format. Tripartite motif-containing protein 32 (TRIM32) is a member of the tripartite motif (TRIM) protein family, characterized by a unique domain structure that includes a B-box, a RING-finger, and a coiled-coil motif, along with six C-terminal NHL domains. TRIM32 is predominantly expressed in skeletal muscle, where TRIM32 plays a crucial role as an E3 ubiquitin ligase, facilitating the attachment of ubiquitin to target proteins in collaboration with ubiquitin-conjugating enzymes such as UbcH5a, UbcH5c, and UbcH6. The ubiquitination process is vital for regulating protein degradation, cellular signaling, and various cellular processes, making TRIM32 essential for maintaining muscle integrity and function. Mutations in the TRIM32 gene are linked to two forms of autosomal recessive muscular dystrophy, namely limb girdle muscular dystrophy type 2H (LGMD2H) and sarcotubular myopathy (STM), as well as Bardet-Biedl syndrome (BBS), which is characterized by a range of symptoms including pigmentary retinopathy, polydactyly, and obesity. TRIM32 (8H8) antibody effectively detects this important protein, making TRIM32 (8H8) antibody a valuable tool for research into these associated disorders and the underlying mechanisms of muscle function and pathology.

For Western Blot, is it recommended to use denatured or non-denatured conditions with TRIM32 (8H8): sc-135588 antibody?

In NSC-34 cell lysate only a non-specific band at ~100kDa is detected but no signal at the expected molecular weight. Incubation O/N at 4 degrees diluted 1:1000 in 5% non-fat dry milk in TBS-T.
