TRA-1-81 Antibody (TRA-1-80) is a mouse monoclonal IgM kappa light chain antibody that specifically recognizes the high molecular weight glycoprotein podocalyxin-like protein 1 (PODXL), also known as PCLP1, gp200, and thrombomucin. The TRA-1-81 antigen is a carbohydrate epitope expressed on podocalyxin, a type I transmembrane glycoprotein. PODXL monoclonal antibody (TRA-1-80), raised against 2102Ep human embryonal carcinoma cells, is reactive with human TRA-1-81 antigen by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). PODXL is predominantly located on the apical membrane of cells, particularly in the kidney′s glomerular podocytes, playing a crucial role in maintaining the filtration barrier, as well as in cell adhesion and migration. PODXL expression on the cell surface is a key marker for undifferentiated human embryonic stem cells and embryonal carcinoma cells. TRA-1-81 expression is lost upon stem cell differentiation, making PODXL monoclonal antibody (TRA-1-80) an important marker for identifying undifferentiated cells. Anti-PODXL antibody (TRA-1-80) is instrumental in identifying pluripotent stem cells in vitro, often used alongside other stem cell markers such as TRA-1-60 and Stage-Specific Embryonic Antigens (SSEA-3 and SSEA-4). PODXL (TRA-1-80) antibody reacts with a high-molecular-mass carbohydrate epitope on the surface of human embryonal carcinoma (EC), embryonic germ (EG), embryonic stem (ES), and induced pluripotent stem (iPS) cells. Anti-PODXL antibody (TRA-1-80) is available in both non-conjugated form and various conjugated forms, including phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different applications. TRA-1-81 antigen detection is essential for stem cell research and for identifying embryonal carcinoma positive nonseminomatous testicular germ cell tumors (NSTGCT), where PODXL is co-expressed with TRA-1-60 and SSEAs on the membrane of numerous stem cells. PODXL monoclonal antibody (TRA-1-80) serves as a valuable tool in stem cell biology and cancer research, facilitating the study of cell surface markers involved in cell differentiation, adhesion, and tumorigenesis.
