TPO Antibody (F-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects TPO protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). TPO Antibody (F-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Thrombopoietin (TPO), also known as c-Mpl ligand, is a crucial cytokine that significantly influences megakaryopoiesis, the process of platelet production from megakaryocytes. TPO is primarily expressed in mature and progenitor megakaryocytes, as well as in human platelets, highlighting its essential role in hematopoiesis. The mature TPO protein is derived from a 332 amino acid precursor, which includes a 21 amino acid signal peptide that is cleaved during processing. TPO exhibits a range of molecular weights in serum, indicating extensive glycosylation, which is vital for stability and activity. TPO interaction with c-Mpl receptor triggers a cascade of intracellular signaling through the activation of Janus kinase family members, including Tyk 2 and JAK2, leading to Stat5 and Stat3 phosphorylation. This phosphorylation facilitates nuclear translocation of these transcription factors, ultimately promoting the expression of genes responsive to Stat signaling. TPO gene is located on human chromosome 3q26.3-q27, underscoring genetic significance in platelet production regulation and overall hematological health.
