SEE ALSO...

TPMT Antibody (E-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects TPMT protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TPMT (E-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Thiopurine S-methyltransferase (TPMT), also known as thiopurine methyltransferase, plays a crucial role in the metabolism of thiopurine drugs, such as 6-mercaptopurine, by catalyzing their S-methylation. This enzymatic activity determines therapeutic efficacy and potential toxicity of these medications, as variations in TPMT activity can lead to significant differences in drug response among individuals. TPMT is primarily located in the cytoplasm and typically exists as a monomer, although S-adenosyl-L-homocysteine can inhibit TPMT activity. Notably, TPMT activity varies across different ethnic groups due to genetic polymorphisms, with a significant proportion of Caucasians exhibiting high TPMT activity. Understanding TPMT function and regulation is essential for optimizing thiopurine therapy and minimizing adverse effects, making anti-TPMT antibody (E-8) a valuable tool for research and clinical applications.

What is the recommended fixation method for immunofluorescence staining with TPMT (E-8): sc-374154 monoclonal antibody?

Asked by: Professor Griffin
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
