TOP3B encodes DNA topoisomerase III beta (Topo IIIβ), an ATP-independent type IA topoisomerase that resolves topological stress by transiently cleaving and rejoining single-stranded nucleic acids. Topo IIIβ functions in genome maintenance pathways that safeguard replication and transcription, and it cooperates with helicase and scaffold partners to limit accumulation of R-loops and other aberrant nucleic acid structures. Through these activities, TOP3B helps preserve DNA integrity and supports proper RNA metabolism, linking topoisomerase activity to both nuclear and cytoplasmic processes. Genetic disruption or altered regulation of TOP3B has been associated with genome instability phenotypes and has been investigated in the context of neurodevelopmental and psychiatric disorder susceptibility.
Topo IIIβ CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the TOP3B gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the TOP3B together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the TOP3B open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Topo IIIβ protein expression.
This CRISPR knockout system enables efficient generation of TOP3B-deficient cell models for investigation of Topo IIIβ signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
