    SEE ALSO...

TopBP1 Antibody (33) is a mouse monoclonal IgG1 antibody that detects TopBP1 in human samples through applications such as western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). TopBP1 is a crucial protein that plays a significant role in the DNA damage response, transcription, and replication processes, containing eight BRCT motifs that are characteristic of proteins involved in these critical cellular functions. Anti-TopBP1 antibody (33) recognizes nuclear foci and localizes to sites of DNA damage or stalled replication forks, underscoring TopBP1′s importance in maintaining genomic stability. Notably, TopBP1 is phosphorylated in response to DNA double-strand breaks and replication blocks, which is vital for function in the DNA damage response pathway. Furthermore, TopBP1 interacts specifically with the C-terminal region of topoisomerase II beta, suggesting facilitation of catalytic reactions by promoting transient breakages of DNA strands. The gene encoding TopBP1 is located on chromosome 3q22.2, and downregulation has been associated with reduced cell survival, likely due to increased apoptosis, highlighting TopBP1′s essential role in cellular health and response to DNA damage.

I am using TopBP1 (33): sc-136106 for immunofluorescence, what dilution of this antibody should I use?

    Asked by: cjMara
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
