Tnp2 encodes transition protein 2 (TNP2), a sperm-specific chromatin remodeling factor that replaces histones during late spermiogenesis and facilitates the subsequent incorporation of protamines for stable DNA compaction. This protein participates in the coordinated histone-to-protamine exchange program, supporting nuclear condensation, genome integrity, and proper sperm head morphology. Disruption of Tnp2 perturbs chromatin packaging dynamics and can lead to impaired sperm maturation and reduced male fertility phenotypes. As a model, mouse TNP2 is widely used to study epigenetic reprogramming, post-meiotic transcriptional shutdown, and the regulation of chromatin architecture in germ cells.
TNP2 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Tnp2 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Tnp2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Tnp2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish TNP2 protein expression.
This CRISPR knockout system enables efficient generation of Tnp2-deficient cell models for investigation of TNP2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
