TNFα Antibody (52B83) is a mouse monoclonal IgG1 kappa light chain antibody that detects TNFα in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), flow cytometry (FCM), and enzyme-linked immunosorbent assay (ELISA). TNFα (52B83) antibody is available in a non-conjugated form, allowing for versatile experimental use. Tumor necrosis factor alpha (TNFα), also known as cachectin, is a critical pleiotropic cytokine that plays a significant role in mediating immune responses and inflammation. TNFα is primarily produced by immune cells such as lymphocytes, neutrophils, and macrophages, and is essential for the regulation of various physiological processes, including apoptosis, cell proliferation, and differentiation. TNFα interacts with TNF receptors on target cells, leading to a cascade of signaling events that modulate immune responses and promote inflammation. What is the dilution ration of TNF-a primary antibody for WB

Does this antibody or do you have an antibody that recognize the N-terminal region of TNF?

Thank you for your question . sc-52746 TNFα (52B83)is raised against the full length of the protein . The binding site has not been characterized. We do not offer a TNFa antibody for the N- terminal.

Rated 4 out of 5 by from Worked ok, although some non specific bindingThe antibody was received as a trial aliquot and stored at +4 °C upon arrival, as recommended. An aliquot was prepared by diluting 1:500 in PBS with 0.05% Tween-20 and 2% BSA. Application: Western blot on murine cell lysates. Experimental protocol: The antibody was used at a 1:500 dilution, as suggested in the datasheet. Standard blocking and washing steps were performed using PBS with 0.05% Tween-20 and 5% non-fat dry milk. Results: The antibody produced a clear band at the expected molecular weight. However, several non-specific bands were also detected across the membrane. This suggests that the antibody may have limited specificity under the tested conditions, or that further optimization of blocking, washing, or antibody dilution is required. Despite this, the sensitivity was sufficient to detect the target protein in 10 ug of total lysate. Additional comments: A high level of non-specific binding was observed, resulting in multiple background bands. We recommend optimizing the blocking and washing steps and testing lower antibody concentrations to reduce non-specific signals.

Rated 1 out of 5 by from BV2细胞的WB无法显示条带2周前买的抗体,我用无论是否使用LPS处理BV2细胞,WB的结果都是没有条带,我的dilution 1:200,loading protein是2.6ug,内参蛋白条带很清晰。除此之外,在santacruz购买的的TNF-α也有同样的问题,两者都没有任何条带。
