SEE ALSO...

TMPRSS2 Antibody (H-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects TMPRSS2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-TMPRSS2 antibody (H-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TMPRSS2, a 492 amino acid multimeric serine protease, plays a crucial role in regulating extracellular matrix components, which is vital for processes such as tumor growth and metastasis. TMPRSS2 (H-4) antibody detects protein predominantly expressed in prostate and kidney of mice, as well as in human small intestine, prostate, colon, stomach, and salivary gland. TMPRSS2 features several important structural domains, including a serine protease domain of S1 family, a scavenger receptor cysteine-rich domain, an LDL receptor class A domain, and a transmembrane domain, which collectively contribute to enzymatic activity and functional versatility. TMPRSS2 exists in both full-length form and cleaved protease domain, with expression upregulated by androgenic hormones, indicating potential involvement in prostate carcinogenesis. Anti-TMPRSS2 antibody (H-4) helps researchers study TMPRSS2′s interactions with various growth factors and activation mechanisms, highlighting significance in both normal physiological processes and pathological conditions.

Would this work with Vero cells? Has it been used in them before?

Asked by: Adam UK
Thank you for your questions. Based on tests on a panel of different cell lysate and tissue samples, TMPRSS2 Antibody (H-4): sc-515727 is recommended for detection of TMPRSS2 catalytic chain of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA and should also detect its target protein in Vero cells if it is present in this cell type.

Dear technical support, Has this antibody been validated using KO or silencing? We've tested 4 different anti TMPRSS2 antibodies so far they all give a band at the right siez but 1) even in lysates from cells known not to express TMPRSS2 2) even in KO!

Dear technical support team, I would like to use your TMPRSS2 (H-4) -PE antibody for flow cytometric analysis. Was this antibody validated for flow cytometry and does it require cell permeabilization?

Asked by: ealeksa2
Thank you for your question. TMPRSS2 Antibody (H-4): sc-515727 was raised against amino acids 296-345 mapping within an internal region of TMPRSS2 of human origin, which correspond to the extracellular domain of the protein. The antibody has not been validated for flow cytometry.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with TMPRSS2 (H-4): sc-515727?

Asked by: Cweed
We recommend using one of our our exclusive Mouse IgG Binding Proteins conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. A complete list of available binding proteins is available on our website here: https://www.scbt.com/scbt/browse/support-products-mouse-igg-binding-proteins/_/N-ecrety
