TLE3 (transducin-like enhancer of split 3) encodes a Groucho/TLE family transcriptional corepressor that binds DNA-associated factors such as TCF/LEF, HES/HEY, and other lineage regulators to modulate gene expression programs during development and tissue homeostasis. By recruiting chromatin-modifying complexes, TLE3 influences Wnt/β-catenin-dependent transcription, Notch effector outputs, and additional differentiation-linked pathways. Altered TLE3 activity has been linked to dysregulated cell fate decisions, proliferation control, and transcriptional reprogramming observed across multiple disease contexts, including cancer biology and metabolic phenotypes. These properties make TLE3 a useful target for dissecting pathway crosstalk and epigenetic control of context-specific transcription.
TLE3 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the TLE3 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the TLE3 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the TLE3 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish TLE3 protein expression.
This CRISPR knockout system enables efficient generation of TLE3-deficient cell models for investigation of TLE3 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
