Thyroperoxidase Antibody (MoAb47) is a mouse monoclonal IgG1 kappa light chain antibody that detects Thyroperoxidase in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Anti-Thyroperoxidase antibody (MoAb47) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Thyroperoxidase (TPO) is a crucial enzyme in the synthesis of thyroid hormones, facilitating the iodination of tyrosine residues in thyroglobulin, which is essential for the production of thyroxine (T4) and triiodothyronine (T3). This process is vital for regulating metabolism, growth, and development in vertebrates. TPO is a 933 amino acid type I transmembrane glycoprotein that not only plays a significant role in hormone synthesis but is also a major autoantigen implicated in autoimmune thyroid diseases, such as Hashimoto′s thyroiditis and Graves′ disease. TPO′s structure includes a large N-terminal myeloperoxidase-like domain, a complement control protein domain, and an epidermal growth factor-like domain, which are important for enzymatic activity and interactions with other proteins. Additionally, TPO mediates the organification and intracellular retention of radioiodide, which can lead to targeted tumor cell death, highlighting potential therapeutic implications. Mutations in the TPO gene can result in goitrous congenital hypothyroidism, a condition characterized by impaired iodide organification, underscoring TPO′s importance in thyroid function and overall endocrine health.
