Thyroperoxidase Antibody (A-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects Thyroperoxidase protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Thyroperoxidase (TPO) antibody (A-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TPO is a crucial enzyme in the synthesis of thyroid hormones, facilitating the iodination of tyrosine residues in thyroglobulin, which is essential for the production of thyroxine (T4) and triiodothyronine (T3). This process is vital for regulating metabolism, growth, and development in vertebrates. TPO′s activity is particularly important because TPO not only catalyzes the iodination reaction but also plays a role in the organification of iodide, which is necessary for hormone synthesis. Additionally, TPO is a significant autoantigen in autoimmune thyroid diseases, where the immune system mistakenly targets TPO, leading to conditions such as Hashimoto′s thyroiditis and Graves′ disease. TPO′s structure includes a large N-terminal myeloperoxidase-like domain, a complement control protein domain, and an epidermal growth factor-like domain, which are essential for enzymatic function and interactions with other proteins. Mutations in the TPO gene can result in congenital hypothyroidism, highlighting TPO′s importance in maintaining thyroid health and function.
