Thymidine Kinase Antibody (C-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects Thymidine Kinase (TK1) of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-TK1 antibody (C-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TK1 is a crucial enzyme that plays a significant role in DNA synthesis by catalyzing the phosphorylation of deoxythymidine, which is essential for the incorporation of deoxythymidine into DNA strands. This function is particularly important during cell proliferation, as TK1 levels rise in anticipation of cell division, making TK1 a valuable biomarker for assessing cellular growth and malignancy. Elevated TK1 levels are often associated with various cancers, especially hematological malignancies, and TK1 measurement can aid in diagnosis, treatment, and monitoring of these diseases. Additionally, TK1 is involved in the mechanism of action of several antiviral drugs, such as azidothymidine (AZT), highlighting TK1′s relevance in therapeutic contexts. TK1 can exist in multiple oligomeric states, including monomers, dimers, trimers, and tetramers, which may influence TK1′s activity and interactions within the cellular environment.

Rated 2 out of 5 by from TK1 reviewIn this experiment, we want to troubleshoot a potential phenotype on the endogenous TK1 levels in presence of two differents "non-TK1 expressors". HEK-293T cells were transfected or not with non-TK1 expressors for 48h. Cells were lysed in RIPA-DOC (+ protease inhibitors) for 30min at 4c and spin 10.000rpm/5min/4c. Proteins quantification was done and 50µg of lysates were migrated on a 12% SDS-PAGE. After 4h transfer on nitrocellulose membrane, membrane were blocked with 5% non-fat milk and probe overnight with the anti-TK1 antibody (C-4 from Santa Cruz; dilution 1/500). Following the O/N incubation, membrane was washed with 1x TBS-T and probe with the corresponding secondary HRP-conjugated Ab. WB was revealed and the corresponding data are shown in the attached photo. The detection signal is very poor and we are almost not able to see the corresponding endogenous TK1 band in the mock and Cond2.
