Themis Antibody (A-11) is a mouse monoclonal IgG2a kappa light chain antibody that detects Themis protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Themis antibody (A-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Themis, which stands for thymocyte-expressed molecule involved in selection, is a 641 amino acid protein that is localized in both the nucleus and cytoplasm and exists as three alternatively spliced isoforms. Themis protein is crucial for T-cell development, as Themis plays a significant role in the regulation of T-cell antigen receptor (TCR) signaling, influencing both positive and negative selection of thymocytes. Themis protein interacts with key signaling proteins such as phospholipase C gamma 1, Emt, and GRB2, which are essential for mediating calcium influx and the phosphorylation of extracellular signal-regulated kinase 1. Notably, Themis undergoes rapid phosphorylation on tyrosine residues following TCR stimulation, highlighting Themis′s importance in T-cell activation and maturation. The gene encoding Themis is located on human chromosome 6q22.33, a region associated with various diseases, including early onset intestinal cancer and other genetic disorders, underscoring the potential implications of Themis in both immune function and disease susceptibility.
