TET1 Antibody (4F4) is a mouse monoclonal IgG2a kappa light chain antibody that detects TET1 protein of human origin by western blotting (WB), immunoprecipitation (IP), and enzyme-linked immunosorbent assay (ELISA). Anti-TET1 antibody (4F4) is available as a non-conjugated format. TET1, also known as tet oncogene 1, LCX, or CXXC6, is a 2,136 amino acid protein primarily localized in the nucleus and features a CXXC-type zinc finger domain, which is crucial for DNA demethylation. This process is vital for regulating gene expression and cellular differentiation, particularly during embryonic development and organogenesis. TET1 is expressed in various tissues, including adult ovary, thymus, and skeletal muscle, as well as in fetal lung, heart, and brain, indicating significant roles in both normal development and potential involvement in diseases such as acute myeloid leukemia (AML). TET1 gene is located on human chromosome 10, a region containing over 1,200 genes and is associated with several genetic disorders, including Charcot-Marie-Tooth disease and multiple endocrine neoplasia type 2. Understanding TET1 function and regulation is essential for elucidating roles in epigenetic modifications and implications in cancer biology.

In the description of the Tet1 antibody a molecular weight of 235 kDa is mentioned but your WB image shows a band between 30-46 K?

Thank you for the question. TET1 (tet oncogene 1) is a 2,136 amino acid protein with the expected molecular weight of approx. 235 kDa to be observed in a standard Western Blot experiment for the endogenously expressed full-length protein. The image on the product sheet refers to the Western blot analysis of human recombinant TET1 protein fragment with a much smaller molecular weight.

For Western Blot, is it recommended to use denatured or non-denatured conditions with TET1 (4F4): sc-293186 antibody?
