TDG Antibody (D-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects TDG protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TDG (D-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. TDG protein plays a crucial role in DNA repair by correcting G/T mismatches that arise from the hydrolytic deamination of 5-methylcytosine to thymine, which is vital for maintaining genomic stability and preventing mutations. Located primarily in the nucleus, TDG not only repairs these mismatches but also interacts with retinoic acid receptors, enhancing their ability to bind to response elements, thereby influencing gene expression. Additionally, TDG undergoes post-translational modifications, such as sumoylation by ubiquitin-like proteins SUMO-1 and SUMO-2/3, which significantly affect enzymatic activity and substrate binding affinity. This modification leads to an increase in enzymatic turnover for G/U substrates while diminishing processing activity for G/T mismatches, highlighting the intricate regulatory mechanisms that govern TDG′s function in DNA repair pathways.

Are there any issues with non-specific staining when using this mouse monoclonal antibody on mouse tissue samples in IF or IHC?

Thank you for your question. This antibody is recommended for use with mouse samples in IF and IHC. To eliminate any potential issues with non-specific binding of the anti-mouse secondary, we recommend using the directly conjugated version of the primary. This antibody is available conjugated to HRP, PE, FITC, AF488 and AF647.

Rated 1 out of 5 by from sc-376652 ImmunofluorescenceI tested the antibody sc-376652 for immunofluorescence. Cells were fixed with PFA 4% and permeabilised with 0,05% triton and sc-376652 was used 1:100. Staining was mild and it did not look to be specific to the nucleus.
