TCR α Antibody (H-1) is a mouse monoclonal IgG2a kappa light chain antibody that detects TCR alpha protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). TCR alpha (H-1) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The T cell antigen receptor (TCR) plays a crucial role in the immune response by recognizing foreign antigens and initiating intracellular signaling that activates T cells, transitioning them from a dormant state to an active one. TCR is a heterodimer composed of either alpha and beta or gamma and delta chains, with approximately 95% of circulating T cells expressing the alpha/beta heterodimer, while the remaining 2-5% express the gamma/delta variant. TCR′s functionality is enhanced by association with CD3 chains and the CD4 or CD8 co-receptors, which are essential for effective signal transduction. T helper cells, characterized by CD4 expression, and T cytotoxic cells, marked by CD8 expression, are vital components of the adaptive immune system. CD4, also known as T4 or Leu 3, is a membrane glycoprotein featuring four extracellular immunoglobulin-like domains, which facilitates the binding of TCR to class II major histocompatibility complex (MHC) molecules presented by antigen-presenting cells. This interaction is critical for the activation of T helper cells, as CD4 enhances the avidity of the TCR-MHC complex, thereby amplifying the immune response.

I am using conjugated TCR alpha for flow staining . I have repeated several times with dilution of 1:100 and 1:20 and have had negative results . seems to me this AB has not been validated. Could you suggest any trouble shooting steps.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with TCR α (H-1): sc-515719?

We recommend using one of our our exclusive Mouse IgG Binding Proteins conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.
