TC-PTP Antibody (D-3) is a mouse monoclonal IgM antibody that detects T-cell protein tyrosine phosphatase (TC-PTP) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TC-PTP is a crucial non-transmembrane enzyme that plays a significant role in cellular signaling pathways, particularly in lymphocyte signaling and hematopoietic homeostasis. TC-PTP exists in two main isoforms, p45TC and p48TC, generated through alternative splicing of its noncatalytic domain. p45TC variant localizes to the nucleus and regulates the activity of the catalytic domain through intramolecular interactions. Upon activation of the epidermal growth factor receptor (EGFR), p45TC exits the nucleus and interacts with key substrates, including EGFR and p52Shc, effectively inhibiting the EGF-dependent activation of phosphoinositide 3-kinase and protein kinase B/Akt pathways. In glioblastoma cells, p45TC suppresses the activation of ERK2 mediated by the deleted epidermal growth factor receptor (DEGFR), thereby reducing tumorigenicity in vivo. Additionally, TC-PTP negatively regulates Janus kinases 1 and 3, and TC-PTP deficiency in mice leads to significant defects in the hematopoietic system, highlighting TC-PTP′s essential role in maintaining immune function and cellular homeostasis. The gene encoding human TC-PTP is located on chromosome 18, underscoring TC-PTP′s evolutionary conservation and importance across species.
