SEE ALSO...

TAP2 Antibody (B-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects TAP2 protein of mouse origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). TAP2 (B-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The transporter associated with antigen processing (TAP) plays a crucial role in immune response as TAP is integral to the major histocompatibility complex (MHC) class I antigen-presenting pathway. TAP is composed of two subunits, TAP1 and TAP2, which form stable dimers that create a translocation pore within endoplasmic reticulum (ER) membranes. This structure facilitates peptide movement from cytosol into ER lumen, where peptides are loaded onto MHC class I molecules for presentation to CD8+ T cells. TAP transporter is characterized by N-terminal transmembrane region that forms translocation pore and cytoplasmic peptide-binding pocket, along with several C-terminal nucleotide-binding domains essential for ATP binding and hydrolysis. These ATP-driven conformational changes are critical for effective peptide transport, underscoring TAP2′s importance in maintaining immune surveillance and response.

Does this antibody also recognize TAP1?

Asked by: demi
Thank you for your question. TAP2 (B-2): sc-515576 is a mouse monoclonal antibody that is specific for TAP2, and does not cross-react with TAP1.

Hi, I'm using TAP2 (G-20) sc-11473 Ab (discontinued???) and I'm having problems because this Ab is recognizing TAP2 defective cells (RMA-S cells). Do you know if this Ab also recognize TAP1?

Asked by: gretica77
Thank you for your question. We do not expect the goat polyclonal antibody, TAP2 (G-20): sc-11473, to cross-react with TAP1, however we have not tested for this cross-reactivity specifically. This is a polyclonal antibody and there is not significant identity shared between the epitope and the TAP1 protein. TAP2 (G-20): sc-11473 is a goat polyclonal antibody that is now discontinued. If you would like to test a free 10 µg sample of our recommended replacement monoclonal antibody, sc-515576: TAP2 (B-2), please contact our Technical Service Department. Please call us toll-free at (800) 457-3801 option 2, e-mail <scbt@scbt.com>, or use the live chat on our website.

What application is the blocking peptide sc-515576 P appropriate for?

Asked by: jenniferc
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization
