TAF4 encodes TAF II p135, a core subunit of the TFIID complex that cooperates with TBP and other TAFs to initiate RNA polymerase II–dependent transcription. Through interactions with sequence-specific transcription factors and chromatin-associated regulators, TAF II p135 contributes to promoter recognition, preinitiation complex assembly, and control of cell state–dependent gene expression programs. TAF4 function is closely tied to chromatin remodeling and transcriptional coactivator networks, linking it to regulation of proliferation, differentiation, and stress-responsive transcription. Dysregulated TFIID/TAF activity and altered transcriptional initiation are frequently studied in the context of oncogenic transcriptional reprogramming and other disorders of gene regulation.
TAF II p135 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the TAF4 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the TAF4 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the TAF4 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish TAF II p135 protein expression.
This CRISPR knockout system enables efficient generation of TAF4-deficient cell models for investigation of TAF II p135 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
