SYP Antibody (SVP38) is a mouse monoclonal IgG1 kappa light chain antibody that detects synaptophysin (SYP) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-SYP antibody (SVP38) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Synaptophysin is an integral membrane protein predominantly located in synaptic vesicles of neurons and endocrine cells, where SYP plays a crucial role in the regulation of neurotransmitter release. SYP forms hexameric complexes, which are essential for maintaining the structure and function of synaptic vesicles. SYP is involved in the endocytosis and recycling of synaptic vesicles, a process vital for sustaining synaptic transmission and plasticity. SYP presence in early endosomes indicates a role in the trafficking and sorting of synaptic vesicle proteins, which is critical for neuronal communication. SYP interacts with other proteins such as synaptobrevin, contributing to the dynamic regulation of synaptic vesicle pools during neuronal development. SYP ability to form complexes with membrane cholesterol further underscores an importance in vesicle formation and function, highlighting significance in the intricate processes of synaptic transmission and neuronal signaling.

Why do you see bands at different molecular weight with WB depending on the cell type? Do you have an explanation for this? thanks

Thank you for your question. Synaptophysin is subject to glycosylation and phosphorylation and has two different isoforms with 33 kDa and 20 kDa, respectively. These factors contribute to the observed molecular weight in western blots based on the cell type they stem from.

Is this antibody suitable for fluorescence immunocytochemistry? I could not find any image showing this.

Thank you for your question. This antibody is recommended for detection of SYP of mouse, rat and human origin by Western Blot, IP, Immunocytochemistry on methanol-fixed cells, Immunohistochemistry on acetone-fixed frozen tissues sections and Immunohistochemistry on paraffin-embedded tissues sections fixed with formalin.
