SYP Antibody (H-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects synaptophysin (SYP) in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-SYP antibody (H-8) is available in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates. Synaptophysin is a crucial integral membrane protein predominantly found in neurons and endocrine cells, where SYP (H-8) monoclonal antibody plays a vital role in the formation and recycling of synaptic vesicles. Synaptophysin is characterized by N-glycosylation and the ability to form hexameric complexes, which contribute to functioning as a large conductance channel. SYP contains four transmembrane domains and regulates membrane cholesterol, essential for vesicle formation. Synaptophysin interacts with synaptobrevin (VAMP), forming a complex upregulated during neuronal development, although not essential for the exocytosis process itself. The tail domain of brain myosin V forms a stable complex with synaptobrevin II and synaptophysin, which disassembles upon calcium ion influx during depolarization, highlighting the dynamic nature of synaptic vesicle cycling and SYP′s role in neurotransmission.

I am using SYP (H-8): sc-55507 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
