SV40 T Ag Antibody (Pab 108) is a mouse monoclonal IgG2a kappa light chain antibody that detects SV40 T Ag of SV40 origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-SV40 T Ag antibody (Pab 108) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SV40 T antigen plays a crucial role in the viral life cycle and is essential for the transformation of host cells, as SV40 T antigen interacts with key cellular proteins that regulate cell growth and division. Notably, SV40 large T antigen binds to the retinoblastoma (Rb) tumor suppressor protein, specifically targeting its "pocket" domain, which is also the binding site for the E2F transcription factor. This interaction leads to the inactivation of Rb, thereby releasing E2F and promoting the transcription of genes necessary for cell cycle progression. I am using SV40 T Ag (Pab 108): sc-148 to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?

Yes, this antibody has been validated for use in IHC with paraffin-embedded tissue sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 5 out of 5 by from LT-Ag - IF & WBLT-Ag antibody worked well on both Methanol:Acetone (50:50) and 10% formalin fixed slides at 1:200. Western blotting was effective at 1:250.

Rated 3 out of 5 by from Good antibody for immunofluorescenceGood antibody for immunofluorescence of SV40LT-transduced cells but unsuitable for WB. I've tested it for WB in several human SV40LT-transduced cell lines (including TC28a2 chondrocytes and immortalized MSC lines) and several non-specific bands were present for all transduced and non-transduced cells.

Rated 2 out of 5 by from Non specific bandsI did a western blot in order to detect the expression of SV40 LT Ag in SV40 LT transfected human cells. I've tried blocking with milk 5% and BSA 3% several times but I am always getting the same result: one specific band of approximately 90 kDa, but accompained by non specific bands (the most intense very near to the specific band of 90 kDa, apppearing in both negative and positive controls). I've tried primary mesenchymal stem cells (MSCs) from several patients as negative controls and T/C28a2 cell line anlong with Phoenix-A cell line (derived from 293T cell line) -both known to express SV40 LT Ag- as positive controls.
