Stim1 Antibody (F-2) is a mouse monoclonal IgG1 antibody that detects Stromal Interaction Molecule 1 (Stim1) in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Stim1 is a crucial cell surface transmembrane glycoprotein that plays a significant role in mediating calcium influx, particularly following the depletion of intracellular calcium stores. Located primarily in the endoplasmic reticulum, Stim1 functions as a calcium sensor due to its EF-hand domain, which undergoes substantial conformational changes in response to fluctuating calcium levels. When intracellular calcium levels drop, Stim1 translocates to the plasma membrane, where Stim1 activates the calcium release-activated calcium (CRAC) channel subunit, TMEM142A/Orai1, facilitating the influx of extracellular calcium. This process is vital for various cellular functions, including secretion and transcription, and highlights the importance of calcium signaling in maintaining cellular homeostasis. Additionally, Stim1′s interaction with Stim2, which acts as a potent inhibitor of Stim1-mediated store-operated calcium entry, underscores Stim1′s regulatory role in calcium dynamics. The implications of Stim1 extend beyond basic cellular functions, as Stim1 has been associated with tumor growth suppression and the interactions between stromal and hematopoietic cells, making Stim1 a significant target for research in cancer biology and cellular signaling pathways.
