Stim1 Antibody (CDN3H4) is a mouse monoclonal IgG1 kappa light chain antibody that detects Stim1 in mouse, rat, and human samples through western blotting (WB) and immunoprecipitation (IP). Anti-Stim1 antibody (CDN3H4) is available in a non-conjugated form, providing researchers with a versatile tool for studying this critical protein. Stim1, or stromal interaction molecule 1, is a vital transmembrane glycoprotein located in the endoplasmic reticulum (ER), where Stim1 functions as a calcium sensor. Stim1′s ability to detect changes in intracellular calcium levels is crucial for various cellular processes, including secretion and gene transcription. Upon depletion of calcium stores, Stim1 undergoes significant conformational changes that enable translocation to the plasma membrane, where Stim1 activates the calcium release-activated calcium (CRAC) channel subunit, TMEM142A/Orai1. This mechanism is essential for maintaining cellular calcium homeostasis and facilitating calcium influx, which is necessary for numerous physiological functions. Additionally, Stim1′s interaction with Stim2, a known inhibitor of Stim1-mediated store-operated calcium entry, highlights Stim1′s regulatory role in calcium signaling pathways. The implications of Stim1 extend to tumor growth suppression and the interactions between stromal and hematopoietic cells, making Stim1 a significant focus of research in cancer biology and cellular signaling.

I am using Stim1 (CDN3H4): sc-66173 antibody for immunoprecipitation, how much antibody should I use?

Asked by: Trav11
Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration. Please find our full immunoprecipitation protocol here: https://www.scbt.com/scbt/resources/protocols/immunoprecipitation-western-blots
