SREBP-1 Antibody (2A4) is a mouse monoclonal IgG1 kappa light chain antibody that detects SREBP-1 protein of mouse, rat, human, and hamster origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry (IHC). SREBP-1 (2A4) antibody is available in both conjugated and non-conjugated forms, providing flexibility for various experimental needs. SREBP-1 plays a crucial role in lipid metabolism, particularly in the regulation of cholesterol levels within cells. SREBP-1 is synthesized as a precursor that resides in the nuclear envelope and endoplasmic reticulum, and when cellular sterol levels are low, SREBP-1 is cleaved to release a soluble fragment that translocates to the nucleus, where SREBP-1 activates the transcription of genes involved in lipid synthesis and uptake, including the low-density lipoprotein (LDL) receptor. This process is vital for maintaining cellular cholesterol homeostasis, as the LDL receptor mediates the endocytic uptake of cholesterol-carrying lipoproteins, thereby controlling cholesterol levels in both cells and plasma. The regulation of SREBP-1 is tightly controlled by sterol levels, as excess sterols inhibit SREBP-1 cleavage, highlighting the importance of SREBP-1 in lipid metabolism and overall cellular health.

I used the sc-13551 antibody to detect SREBP1 expression in the kidneys and livers of diabetic rats, with a sample loading amount of 64 μg. I detected the protein in the liver but not in the kidney. How should I adjust the experiment?

Asked by: Vinco1076
Thanks for your question. SREBP-1 is expressed predominantly in brown adipose tissue. The positive results in rat liver indicate that there is no problem with your experimental protocol and antibody. It is recommended that you refer to the literature to confirm the expression of this protein in rat kidney tissue.

What is the difference between sc-13551 and sc-13551X? Can the sc-13551: mouse monoclonal SREBP-1 Antibody (2A4) be used for Supershift Assay?

Asked by: Taro
Thank you for your question. The only difference between these two products is that the “X” form is 10x more concentrated and vialed in a buffer without stabilizing protein (gelatin). This TransCruz reagent “X” is for Gel Supershift (EMSA) and ChIP applications. sc-13551 X, 200 µg/0.1 ml
