SRC-1 Antibody (1135/H4) is a mouse monoclonal IgG1 kappa light chain antibody that detects SRC-1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-SRC-1 antibody (1135/H4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SRC-1 protein, also known as steroid receptor coactivator-1, plays a crucial role in the regulation of gene expression by acting as a coactivator for nuclear hormone receptors, which are essential for mediating the effects of steroid hormones, thyroid hormones, and retinoic acids. SRC-1 is primarily located in the nucleus, where SRC-1 interacts with transcription factors to enhance the transcription of target genes. Nuclear localization of SRC-1 is vital because SRC-1 allows for precise regulation of gene expression in response to hormonal signals, thereby influencing various physiological processes such as metabolism, development, and reproduction. SRC-1 undergoes post-translational modifications, including phosphorylation, which can modulate activity and interactions with other proteins, further emphasizing SRC-1′s importance in cellular signaling pathways.

Hi , I have ordered this antibody for protein src ( sc-32789), why it's molecular weight is so high (Mol.wt- 160 Kda), as compared to molecular weight of its phosphorylated form (p-src mol wt- 60 Kda). I have been looking for total protein form of p-src

Thank you for your question. This antibody is specific for SRC-1 (for steroid receptor coactivator-1, also designated NCoA-1). This protein has a predicted molecular weight of 160 kDa. You are comparing this to a different protein/gene, p-src, or p-c-Src, which is part of the Src gene family of membrane-associated tyrosine protein kinases. If you are looking for a total antibody against this latter target, then I would recommend a different antibody, c-Src Antibody (B-12): sc-8056.

When should the TransCruz reagent sc-32789 X be used instead of the standard form sc-32789?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
