SQSTM1 Antibody (A-6) is a mouse monoclonal IgG1 kappa kappa light chain antibody engineered to specifically detect SQSTM1, also known as p60, p62, A170, DMRV, OSIL, PDB3, ZIP3, p62B, EBIAP, NADGP, and FTDALS3, across multiple species including mouse, rat, and human. Anti-SQSTM1 antibody (A-6) targets the amino acid region 151-440 of the human SQSTM1 protein in applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). SQSTM1 is a multifunctional adaptor protein involved in various cellular processes, including autophagy, where SQSTM1 facilitates the selective degradation of ubiquitinated proteins by interacting with LC3 on autophagosomes. Additionally, SQSTM1 interacts with key signaling proteins such as TRAF6, which plays a pivotal role in the NF-κB signaling pathway, and Keap1, which regulates the Nrf2 pathway involved in oxidative stress responses. These interactions are crucial for maintaining cellular homeostasis and regulating inflammation, apoptosis, and oxidative stress responses. SQSTM1′s binding to ubiquitinated proteins is further modulated by post-translational modifications, including phosphorylation and ubiquitination, which enhance binding affinity and functional versatility in signaling pathways and autophagic flux. SQSTM1 (A-6) antibody is available in both non-conjugated form and conjugated forms such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing researchers with versatile options for diverse experimental needs. Anti-SQSTM1 antibody (A-6) is instrumental in studying pathological conditions like Paget′s disease of bone, where mutations in the ubiquitin-associated (UBA) domain of SQSTM1 disrupt normal function in aggregate sequestration and cell survival, leading to abnormal bone remodeling and increased bone fragility. SQSTM1 monoclonal antibody (A-6) enables researchers to gain comprehensive insights into molecular interactions and regulatory mechanisms of SQSTM1, facilitating advancements in understanding disease mechanisms and developing targeted therapeutic strategies.
