
SPT5 Antibody (E-8) is a mouse monoclonal IgG1 antibody that detects SPT5 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SPT5, also known as DSIF p160, plays a crucial role in the regulation of transcriptional elongation, particularly in the context of RNA polymerase II activity. SPT5 is primarily located in the nucleus, where SPT5 forms a complex with SPT4 to create the DRB-sensitivity-inducing factor (DSIF) complex, which is essential for modulating the transcription process. The interaction between SPT4 and SPT5 is vital for the recruitment of RNA polymerase II, thereby influencing the efficiency of gene expression. Notably, SPT5 undergoes post-translational modifications, including phosphorylation, which significantly alters mobility on SDS-PAGE and enhances regulatory functions. The C-terminal CTR1 domain of SPT5 is specifically phosphorylated by the P-TEFb complex, a modification that is critical for transcriptional elongation.
