Somatostatin Antibody (H-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects Somatostatin in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Somatostatin antibody (H-11) is available in both non-conjugated and multiple conjugated forms, such as agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and various Alexa Fluor® conjugates. Somatostatin is a crucial regulatory hormone that plays a significant role in inhibiting the release of several secondary hormones by binding to high-affinity G protein-coupled somatostatin receptors. This cyclic tetradecapeptide inhibits the secretion of key hormones including growth hormone, insulin, and glucagon. Somatostatin is predominantly located in the hypothalamus and pancreas, where Somatostatin regulates insulin synthesis and overall metabolic processes. Somatostatin exists in two active forms—Somatostatin-14 and Somatostatin-28—produced through the cleavage of a single preproprotein, with distinct expression patterns in the cerebellum at different developmental stages. The gene encoding Somatostatin is located on human chromosome 3q27, highlighting Somatostatin′s importance in both normal physiological functions and potential implications in tumorigenesis and neurotransmission within the central nervous system.

Can I use SST Ab(H-11) on mouse? I saw some non-specific binding when I did the IHC with SSt Ab(H-11) on mouse brains

Can sc-74556 label for both ss-14 and ss-28 on a western blot? ie would it recognise both forms that could be viewed by a difference in size?

Thank you for your question. The expected molecular weight for this protein is ~17kDa. We do not have information about seeing the 28kDa band.

Can Somatostatin (H-11): sc-74556 antibody be used to stain free floating thick (50µm) vibratome mouse brain slices, fixed overnight with 4% PFA? If so, does it require antigen retrieval? We need to use it with a fluorescent secondary antibody.

Thank you for your question. We have not tested this antibody specifically on free floating tissue sections. However, we do recommend the antibody for immunohistochemistry on formalin-fixed tissue sections. If you are using 4% PFA for fixation, we do recommend antigen retrieval. Our preferred method is sodium citrate buffer (pH 6.0) with heat treatment, but that may not be suitable for your free floating tissue sections. We would recommend testing different antigen retrieval methods with the antibody in your application to determine the optimal results.

Can Somatostatin (H-11): sc-74556 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, Somatostatin (H-11): sc-74556 has been validated for use in IHC with paraffin-embedded sections. Rated 4 out of 5 by from Good signal, low background, standard protocolI tried this as a sample and I can recommend it. I used standard protocol for DAB staining and it worked right away. I did not use any amplification kit, just very basic chemicals and DAB in powder. So I believe the signal still can be improved a lot if protocol is optimized. Protocol notes: - antigen retrieval in citrate buffer for 10 minutes - 30 min pre-incubation with 1% H2O2 - blocking for 1 h in PBS + 0.15% Triton + 5% BSA + 5% goat serum - incubation with primary o/n (in PBS + 0.15% Triton + 2% goat serum) - incubation with secondary 2 hrs in the same solution - washing always in PBS + 0.15% Triton
