SNRPC encodes a core Sm protein component of the U1 small nuclear ribonucleoprotein (snRNP), an essential subcomplex of the spliceosome that recognizes 5′ splice sites and promotes accurate pre-mRNA splicing. Through assembly of snRNP particles and stabilization of U1 snRNA–protein interactions, SNRPC supports spliceosome biogenesis, co-transcriptional RNA processing, and downstream gene expression programs. Disruption of spliceosomal components, including SNRPC, is linked to widespread splicing dysregulation that can affect cell-cycle control, DNA damage responses, and differentiation pathways. Altered spliceosome function has been associated with diverse human diseases, particularly cancers and inherited disorders driven by aberrant RNA processing.
SNRPC CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the SNRPC gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the SNRPC together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the SNRPC open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish SNRPC protein expression.
This CRISPR knockout system enables efficient generation of SNRPC-deficient cell models for investigation of SNRPC signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
