SmcX Antibody (C-12) is a mouse monoclonal IgG2b antibody that detects SmcX in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SmcX, also known as JARID1C, MRXJ, KDM5C, or XE169, is a crucial nuclear protein that plays a significant role in the regulation of gene expression and chromatin dynamics. SmcX contains one ARID domain, one JmjC domain, one JmjN domain, and two PHD-type zinc fingers, classifying SmcX within the JARID1 histone demethylase family. SmcX is predominantly expressed in the brain and skeletal muscle, where SmcX functions as a histone demethylase, specifically removing methyl groups from lysine residues on Histone H3. This demethylation is vital for maintaining the histone code, which in turn influences transcriptional regulation and chromatin remodeling. SmcX′s ability to bind iron and α-ketoglutarate as cofactors enhances enzymatic activity, allowing SmcX to recruit histone deacetylases to neuron silencer elements, thereby repressing the transcription of neuronal genes. Importantly, defects in the gene encoding SmcX are linked to X-linked mental retardation (XLMR), a condition that results in cognitive impairment and low IQ. Additionally, multiple isoforms of SmcX arise from alternative splicing, contributing to functional diversity in cellular processes.
