Six1/2/4 Antibody (E-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects Six1/2/4 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Six1/2/4 (E-11) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Six1/2/4 protein plays a crucial role in vertebrate development and tissue differentiation, functioning as a transcription factor that binds to specific DNA motifs to regulate gene expression. Notably, Six1 protein is over-expressed in many primary mammary carcinomas and metastatic lesions, highlighting its importance in cancer biology. Additionally, Six1 and Six4 are involved in muscle formation by activating the regulatory protein myogenin through binding to the MEF3 DNA motif, which is essential for proper muscle development. Human SIX1 gene is located on chromosome 12q23 and is expressed as a nuclear phosphoprotein in adult skeletal muscle, while SIX4 gene, found on chromosome 14q23, encodes a 781 amino acid protein expressed in the developing brain and spinal cord. Human SIX2 gene, mapping to chromosome 2p16-p15, is also expressed in skeletal muscle, pancreas, ovary, and sclera, further emphasizing the significance of the Six gene family in various biological processes.
