SIT Antibody (G-8) is a mouse monoclonal IgG1 antibody that detects SIT in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SIT, or SHP2 interacting transmembrane adaptor protein, plays a crucial role in T lymphocyte signaling by acting as a disulfide-linked homodimeric glycoprotein that facilitates the recruitment of SH2 domain-containing intracellular signaling molecules to the plasma membrane via tyrosine-based signaling pathways. The presence of five potential tyrosine phosphorylation sites on SIT indicates its importance in T cell receptor-mediated signaling, which is vital for effective immune responses. Additionally, SIT interacts with key proteins such as SHP2 and the adaptor protein Grb2, and serves as a substrate for Src family kinases, including Fyn, Lck, and ZAP-70, which are essential for T cell activation and function. The ability of SIT to undergo endoglycosidase treatment, resulting in a reduction of its molecular mass, further highlights its dynamic role in cellular signaling processes. Anti-SIT antibody (G-8) is an invaluable tool for researchers studying T cell signaling pathways and their implications in immune responses.
