SHP Antibody (H-5) is a mouse monoclonal IgG2a kappa light chain antibody that detects SHP protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). anti-SHP antibody (H-5) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SHP, also known as small heterodimer partner, is an orphan nuclear receptor that plays a crucial role in regulating metabolic processes by modulating the activity of other nuclear receptors. SHP lacks a DNA binding domain, which distinguishes SHP from other nuclear receptors, allowing SHP to function primarily as a negative regulator. SHP is predominantly expressed in the liver, fetal liver, adrenal gland, spleen, and small intestine, highlighting SHP′s importance in metabolic regulation and homeostasis. SHP interacts with various nuclear receptors, including thyroid receptors, retinoic acid receptors, and estrogen receptors, inhibiting their activity through mechanisms such as dimerization and competition for coactivators. SHP is involved in the transcriptional regulation of NF-κB in macrophages, suggesting SHP′s role as a modulator in inflammatory responses. SHP regulation is critical for maintaining bile acid homeostasis, as SHP is induced by bile acids through the farnesoid X receptor (FXR), providing a protective mechanism against bile acid-induced hepatocyte damage in cholestatic conditions.

Can this antibody detect a truncated SHP which is only 1-52 aa? Has the epitope been mapped? I am looking for one that can detect the truncated mutant. Thanks.

Is there anti-shp for mouse?

Thank you for your question. We currently offer 3 different mouse monoclonal antibodies against SHP, and all 3 are recommended for detection of SHP of human origin. We have not yet confirmed reactivity with mouse or rat SHP for these antibodies. The antibodies are raised against a recombinant protein that does share 77% sequence identity with the mouse SHP protein, so there is a very good chance that they will also be reactive with mouse. For a limited time, we are offering a free 10 µg sample of our mouse monoclonal antibody, SHP (H-5): sc-271511. Can SHP (H-5): sc-271511 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, SHP (H-5): sc-271511 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
